The role of mitochondria in cancer was the central theme of the congress held in mid-September in Prague, Czech Republic, where researchers from the Center for Integrative Biology (CIB) of the Universidad Mayor, Julio César Cárdenas and Alenka Lovy participated.

The Doctors presented their work on the regulation of mitochondria through calcium and what is its impact on tumor growth.

My presentation was a particular group of tumors that shows a special sensitivity to this regulation. While in the vast majority of tumors, spontaneous Ca2 + signaling from the intracellular Ca2 + release channel of InsP3R to mitochondria is essential for optimal oxidative phosphorylation (OXPHOS) activity and ATP production, which cancer cells use to grow, migrate and metastasize, there is a group of tumors and tumor cells in which the cells have a defective OXPHOS or do not use it given environmental conditions (for example hypoxia). In this case, the cells replace OXPHOS with reductive carboxylation to maintain levels of reductive equivalents and metabolic precursors. If mitochondrial calcium plays a role in the regulation of the bioenergetics of these cells, it was a mystery until the development of this work, ”explained Cárdenas.

The researcher added that “using OXPHOS-competent and defective cells, we demonstrate that inhibition of InsP3R activity or capture of Ca2 + by mitochondria is required for an optimal activity of αKG-dehydrogenase (αKGDH) necessary for reductive carboxylation. Inhibition of αKGDH activity and the concomitant elevation of NAD + induce cell death in these cells. These results demonstrate that mitochondria play a fundamental role in maintaining bioenergetic homeostasis of both competent and defective cells for OXPHOS. The inhibition of the transfer of Ca2 + from ER to mitochondria represents a new therapeutic strategy to eliminate cancer cells regardless of their OXPHOS status."

For its part, the assistant professor of the CIB showed how mitochondrial dynamics, that is, the way this organelle is found, affects tumor growth.

"Mitochondria are a highly dynamic interconnected network of organelles in constant fusion and fission, which allows the mixing of metabolites and mitochondrial DNA to ensure adequate quality and performance. Ca2 + regulates many aspects of mitochondrial physiology by modulating the activity of several mitochondrial proteins Previously we showed that inhibition of the transfer of Ca2 + mediated by IP3R to mitochondria leads to metabolic stress that eventually leads to the death of tumor cells Here we show that the decrease in mitochondrial function generated by the lack Ca2 + transfer induces mitochondrial fragmentation that is mediated at an early stage by an increase in the NAD + / NADH ratio and the activation of SIRT1, which acts independently of Drp1. Activation of SIRT1 leads to the deacetylation of cortctin, which favors actin polymerization and mitochondrial fragmentation. This dates reveal for the first time that SIRT1 is a new player in the regulation of mitochondrial fragmentation induced by metabolic / bioenergetic stress".

For researchers it was a great experience, considering the exchange with international experts.

“Everything we learned is going to help us now publish our work in international journals and that is the importance of having participated: generating this feedback and being in contact with top-level researchers, showing them what we are capable of doing in Chile and what we do quality research at the University, ”said Dr. Cárdenas.
